ABSTRACT

OBJECTIVE@#To observe the change of the specificity of the microcirculatory blood perfusion at the area of "Feishu" (BL 13) in the rats of chronic obstructive pulmonary disease (COPD).@*METHODS@#According to the random number table, 60 Wistar rats were divided into a 29 d model No. 1 group (C1 group), a 29 d normal control No.1 group (N1 group), a 89 d model No.2 group (C2 group) and a 89 d normal control No. 2 group (N2 group), 15 rats in each one. In the C1 and C2 groups, the smoking and intratracheal drops of endotoxin were used in combination to prepare COPD model. The rats were fed normally in the N1 and N2 groups. "Feishu" (BL 13), "Xinshu" (BL 15), the lateral site of "Feishu" (BL 13) and the lateral site of "Xinshu" (BL 15) were selected as the monitoring points. The pericam perfusion speckle imager (PeriCam PSI System) was adopted to monitor the microcirculatory perfusion unit (PU) at the monitoring points before and in 29 d and 89 d after modeling separately.@*RESULTS@#Before modeling, the differences in PU were not significant at each monitoring point in comparison among the 4 groups and the differences were not significant among "Feishu" (BL 13) and "Xinshu" (BL 15) as well as their lateral sites (all >0.05). After modeling, PU was increased at each monitoring point in the C1 and C2 groups (all <0.05). PU in the C1 group was higher than the N1 group and that in the C2 group was lower than the N2 group, PU at each monitoring point in the C1 group were higher than the C2 group, indicating the significant differences (all <0.05). In the C1 and C2 groups, the specific change occurred, in which PU at "Feishu" (BL 13) was higher than its lateral site. But such specific change did not happen in the N1 and N2 groups.@*CONCLUSION@#PU at "Feishu" (BL 13) presents the specific change relevant with the sickness duration in the COPD rats.

ABSTRACT

Mutual promotion, mutual assistance and mutual inhibition are commonly for the compatibility of Chinese herbs, but they are also existed among the acupoints. In the paper, the relevant literature on acupoint, compatibility was collected and the mutual promotion, mutual assistance and mutual inhibition relationships of acupoints were described. The laws of the above mentioned combinations, the theoretic evidences and modern researches were summarized briefly. The mutual promotion and mutual assistance refer to the coordination of acupoints, presenting the relationship of the primary and the secondary about the mutual assistance. Hereby, the acupoints could be divided into the simple assistant acupoints and the acupoints from the affected meridians. The mutual inhibition is used to describe the antagonism effect in acupoint combination. There are more researches on the mutual promotion of acupoint combination but less ones on the mutual inhibition and its mechanism is just hypothetic. It is very important to have the deep study on those compatibility relationships for the improvement of acupuncture efficacy and to elaborate the effect mechanism.

ABSTRACT

Objective To study the effect of dipsacus on the expression of β-amyloid in parietal cortex and hippocampus of Alzheimer's disease model rats.Methods A total of 40 rats were randomly divided into control group,model group,dipsacus group and vitamin E group (n=10,each)General situation of rats was observed after 42 days of dipsacus treatment.The ability of learning and memory in rats was tested by Morris water maze.The content of β-amyloid in parietal cortex and hippocampus of each group rats were measured using double antibody sandwich method.Results Model group compared with control group showed that general conditions deteriorated markedly;escaping delitescence was prolonged significantly(P<0.01) ; the activity time on the former platform quadrant reduced significantly (t=4.6261,P=0.0002) ; the number of crossing the former platform reduced significantly (t=6.5335,P=0.0000) ; the content of Aβ in parietal cortex and hippocampus increased significantly (t=4.2812,P=0.0004) (t=5.2499,P=0.0001).Dipsacus group and vitamin E group compared with model group showed that general condition improved significantly;escape latency was shortened significantly (P< 0.01); the activity time on the former platform quadrant increased significantly [(45.76±12.15) s,(48.70±10.25) s and (30.20±10.48) s (t=3.0666,3.9908,P=0.0066,0.0009)]; the number of crossing the former platform increased significantly [(3.02±1.19) t/2min,(3.56±0.85) t/2min and (1.43 ± 1.24) t/2min (t=2.9256,4.4804,P=0.0090,0.0003)]; the content of Aβ in parietal cortex and hippocampus reduced significantly in parietal cortex [(280.37-51.40) pg/g,(263.14 ± 45.52) pg/g versus (337.46 ±70.51) pg/g (t=2.1164,2.8003,P=0.0485,0.0118)],in hippocampus [(295.60±67.58)pg/g,(274.38±57.56)pg/g versus (388.26±83.72)pg/g(t=2.9256,4.4804,P=0.0090,0.0003)].Conclusions Dipsacus can reduce expression of Aβ in brain areas related to general intelligence that may have anti-Alzheimer's disease action.The active ingredients of dipsacus may be natural vitamin E.

ABSTRACT

BACKGROUND: It has become a hot issue to treat malignant tumors with retroviral mediated cytokines. But it remains a controversy whether retroviral mediated γ-interferon(IFN-γ) is valid in killing glioma cells and prolonging the survival of tumor carriers.OBJECTIVE: To observe the effect of retrovital mediated γ-interferon on the growth of glioma and the survival of tumor-carrying rats.DESIGN: Completely ramdomized trial based on animals.SETTING: Neurosurgery department of a hospital affiliated to a university.MATERIALS: The trial was conducted in the Neurosurgery Institute of Harbin Medical Unicersity from July 2002 to July 2004. The subjects were 60 male Wistar rats provided by the Animal Center of the First Affiliated Hospital of Harbin Medical Unicersity. They were randomly assigned into 3groups: C6 control group, C6/PA317neo control group and C6/PA317IFN-γ treatment group, 20 in each group.INTERVENTIONS: Retroviral vector containing murine IFN-γ gene was transferred into retroviral package cell PA317. A high titer PA317IFN-γ cell line was obtained after G418 resistance screening. In the right basal ganglion area 1 × 106 C6 cells was injected in each of the rats. The rats in C6groups received no more intervention. Those in C6/PA317neo control group received injection of 1 × 106 PA317IFN-γ cells in the right basal ganglion 4days after C6 injection. Those in C6/PA317IFN-γ treatment group received injection of 1 × 106 PA317IFN-γ cells in the right basal ganglion 4 days after selected from each group to be kill, and the brains were taken out to make frozen sections for SABC immunohistochemical staining. The CD4 and CD8 T was measured with imaging processing software under microscope.RESULTS: The mean survival time of rats in C6 and C6/PA317neo control groups was(27.10 ± 0.82) days and (26.60 ± 0.93) days respectively( P > 0.05), while that in treatment group was(49.30 ± 4. 17) days and there were five rats outlived beyond 60 days( P < 0.01 vs the two controls). The tumor volume in the two control groups was of no difference but the tumor volume in treatment was significantly smaller than that in control groups( P < 0. 01 ) . The CD4 and CD8 lymphocytes infiltration in treatment group was more than that in control groups( P < 0.01) and there was no difference between the two controls( P > 0.05) . It was also revealed through immunohistochemical examination that in treatment group there was Brdu positive cells on the 7th day and they disappeared on the 16th day. But there was no Brdu positive cells in the control groups.CONCLUSION: Intratumor injection of PA317IFN-γ induces anti-tumor immunal reaction in rats. Retroviral mediated IFN-γ inhibits tumor growth and prolongs rat survival.
